This college was established by the Catholic Church Sisters' Organization in the year 1934 with the aim of educating girls to increase the number of female teachers in the country. The center used at that time to conduct these training sessions was Kilema Mission where they were taught subjects of Knowledge and Teaching.
The graduating students underwent their examinations at the Singachini Teachers College. Due to the small size of the Kilema mission area, from 1957 to 1959, the college was relocated to a new area in Mandaka and named Mandaka Teachers College. The Assumption Sisters of the Catholic Church were the ones who initiated this college and led it from 1934 to 1970.
In July 1970, the college was taken over by the government and given registration number C.U.11 to continue providing Grade IIIA Teacher Training, and by 1978 the college began offering Certificate level Science Teacher Training for in-service teachers for a period of one year. In 1984, the Grade IIIA course that was being offered was changed to Grade IIIB.
Additionally, in 1993, the college began offering Grade IIIA Training, and in 1996, it began offering a Diploma in Home Economics Education for Grade IIIA teachers with qualifications for two years. From 1934 to 2013, Mandaka Teachers College only prepared female teachers. However, in 2013, the college began preparing both male and female students and has the capacity to enroll 700 students.
